2nd prize: Visualising GPR88-Venus cell distribution, contrasting with astrocytes

Gpr88 is an orphan GPCR enriched in the striatum and involved in dopaminergic behaviours such as locomotion making it a potential target for neuropsychiatric treatments for diseases such as schizophrenia. In this experiment, the fluorescent receptor knock-in mouse, -Gpr88 Venus/Venus allows us to examine receptor distribution at a regional level. This has revealed high levels of expression in the somatosensory cortex as seen in the presented image, which was the result of immunohistochemistry examining the cell subtype of Gpr88 expressing cells. GPR88-Venus is exclusively expressed in neurons, and shows no colocalization with GFAP, a glial antibody and marker of astrocytes, as seen in this image.
Specimen and labels:
Gpr88 Venus/Venus mice brains were coronally sliced. The tissue were then stained with DAPI, Primary antibodies Rabbit Anti-GFP with secondary alexa fluor 488 (Labelled GPR88), and Chicken Anti-GFAP with secondary alexa fluor 594 (Astrocytes).
Image acquired on Olympus IX73 inverted fluorescent microscope at 10x magnification. Channels used are DAPI (Blue), GFP (Green – GPR88) and CY3 (Red - GFAP).
