Reactive astrogliosis at different stages of Alzheimer’s disease –relationship with other pathology biomarkers and clinical implications

Astrocytes are well known to play an important role in the optimal functioning of the brain including regulating the synapse function. The astrocytes respond to changes in brain homoeostasis by a defence process named reactive astrogliosis, which is observed in AD as well in other neurodegenerative disorders. Reactive astrocytes are closely related to amyloid plaque and tau deposition in AD and more recently new data suggest that reactive astrogliosis in the early time course of AD may precede other pathological hallmarks of AD. Translational in vivo /in vitro tracer imaging studies suggest ” a first and a second wave” of the reactive astrogliosis in AD with different relationship with other AD pathology biomarkers at the different stages of the disease. Several new PET tracers for visualizing reactive astrogliosis in brain will provide further valuable insight to disease mechanisms for both AD and other non-AD dementia. The clinical value of new fluid astrocytes biomarkers is a new avenue to be explored.
